Blue/White Cloning of a DNA Fragment & Assay of ß-galactosidase
Students perform DNA subcloning in the pUC polylinker region, disrupting β-galactosidase and producing white colonies among blue. Using a DNA fragment, linear plasmid, and T4 DNA Ligase, they ligate, transform E. coli, and count recombinant colonies. Blue and white colonies are assayed for β-galactosidase activity. The experiment includes three modules: ligation, transformation, and enzyme assay.
